The MYC proto-oncogenes are among the most commonly activated proteins in human cancer. N-myc is primarily implicated in high-grade neuroendocrine tumors (neuroblastoma and small-cell carcinomas) and those arising from the neural system (neuroblastoma, medulloblastoma, and glioblastoma).
It is a potent developmental regulator that controls proliferation, metabolism, ribosome biogenesis, and cell-cycle progression during embryogenesis—especially in neural crest–derived tissues.
Overexpression is generally associated with a more aggressive clinical course and lower survival rates.

MB, Medulloblastoma: Click to Expand ⟱
Medulloblastoma is an aggressive primary brain tumor that arises in the cerebellum of children and young adults.
Activation of these three pathways Akt/NF-kB, Shh, and Wnt/b-catenin converge in the upregulation of C-myc, N-myc, and Cyclin D1, three oncoproteins that play key roles in the development of medulloblastomas. High levels of these three oncoproteins were considered to be bad prognosis because they are related to unfavorable therapeutic outcome.
