| SFXN2 - Sideroflexin 2 Type: Mitochondrial transmembrane protein / sideroflexin family member / mitochondrial iron-regulation protein Function: SFXN2 is a mitochondrial membrane protein involved in mitochondrial iron homeostasis, heme biosynthesis, mitochondrial respiration, redox regulation, and mitophagy. Loss of SFXN2 can cause mitochondrial iron accumulation, reduced heme content, impaired heme-dependent enzyme activity, and mitochondrial respiratory dysfunction. Cancer: ↑ Increased SFXN2 has been associated with tumor-supportive mitochondrial metabolism in selected cancers. In multiple myeloma, SFXN2 is elevated and promotes proliferation while suppressing starvation-induced autophagy and mitophagy. SFXN2 inhibition or knockdown disrupts mitochondrial iron metabolism, increases mitochondrial damage and mitophagy, and suppresses tumor growth. Increased SFXN2 has also been reported in prostate cancer, although its cancer role remains tissue-dependent and less broadly characterized than many established oncogenic targets. |
