Vanillic Acid - 4-Hydroxy-3-Methoxybenzoic Acid

Type: Phenolic acid / hydroxybenzoic acid derivative

Sources: Vanillic acid occurs naturally in numerous plants and foods and is also produced through metabolism of other phenolic compounds including vanillin and ferulic-acid-related compounds.

Function: Vanillic acid is a naturally occurring phenolic acid with antioxidant, anti-inflammatory, metabolic, and antiproliferative activities. It can influence oxidative stress, mitochondrial function, apoptosis, cell-cycle regulation, inflammatory signaling, and pathways including NF-κB, PI3K/AKT/mTOR, JAK/STAT, and MAPK signaling.

Cancer: Preclinical studies demonstrate antiproliferative, pro-apoptotic, anti-invasive, anti-migratory, and anti-angiogenic effects. Reported mechanisms include inhibition of NF-κB, PI3K/AKT/mTOR and JAK/STAT signaling, reduction of VEGF, MMP-2 and MMP-9, cell-cycle arrest, and modulation of mitochondrial apoptosis and oxidative stress. Clinical anticancer efficacy has not been established.

Alzheimer's Disease: Preliminary and indirect evidence suggests potential neuroprotective relevance through antioxidant, anti-inflammatory, mitochondrial, and anti-apoptotic mechanisms. Vanillic acid occurs among bioactive constituents of medicinal plants investigated in Alzheimer's disease models, but direct evidence for vanillic acid as an Alzheimer's treatment remains limited.

AChE is an enzyme that rapidly hydrolyzes the neurotransmitter acetylcholine into choline and acetate, terminating cholinergic signals.
- In some cancers, studies have reported reduced AChE activity, which may contribute to an accumulation of acetylcholine.
- Lower levels or loss of AChE expression/activity have been associated with more aggressive tumor behavior and poor prognosis, possibly due to unchecked cholinergic signaling.

For AD (Alzheimer's), AChE inhibitors are used, to allow ACh, and ChAT to increase along with acetyl-CoA
-Natural AChE inhibitors: Ferulic Acid, Caffeic Acid, Rosmarinic Acid, Sage
-AChE inhibitors only temporarily relieve some of the disease’s cognitive symptoms and do not stop the patient’s cognitive loss
-adverse effects such as disorientation, falls, dizziness, and fatigue may occur with these medications and should be used only as recommended
