| Citrulline - L-Citrulline Abbreviation: Cit, L-Cit Type: Non-protein amino acid / metabolite / urea-cycle intermediate / arginine precursor Function: Citrulline is a non-protein amino acid involved in the urea cycle and arginine metabolism. It can be converted with aspartate to argininosuccinate by ASS1 and subsequently to arginine by ASL. Citrulline therefore contributes to endogenous arginine synthesis and indirectly influences nitric oxide production, polyamine metabolism, and cellular nitrogen balance. Cancer: ↕ Context-dependent. Citrulline is closely linked to tumor arginine metabolism. Many cancers have reduced or absent ASS1 expression and therefore have limited ability to convert citrulline into arginine, contributing to dependence on extracellular arginine. Arginine-depletion therapies such as pegylated arginine deiminase convert arginine to citrulline and can therefore increase circulating citrulline while selectively stressing ASS1-deficient tumor cells. Alzheimer's Disease: ↑ Circulating citrulline has been reported to be increased in Alzheimer's disease and dementia and may reflect altered arginine, nitric-oxide, and urea-cycle metabolism. Increased citrulline has also been reported across the progression from cognitively healthy subjects through mild cognitive impairment to Alzheimer's disease. |
