Type: Polyamine biosynthesis enzyme / pyridoxal phosphate-dependent decarboxylase

Function: Catalyzes the decarboxylation of ornithine to putrescine, the rate-limiting and committed step in polyamine biosynthesis. ODC1 regulates production of putrescine, spermidine, and spermine and thereby influences cell proliferation, growth, differentiation, apoptosis, metabolic signaling, and cellular stress responses.

Cancer: ↑ Frequently increased or overactivated in cancer and is a major downstream target of MYC signaling. Elevated ODC1 increases polyamine synthesis and can promote proliferation, tumor growth, angiogenesis, migration, invasion, and survival. ODC1 inhibition, including with DFMO/eflornithine, suppresses proliferation and tumor growth in multiple experimental cancer models.

Alzheimer's Disease: ↑ Increased astrocytic ODC1 activity is associated with Alzheimer's disease pathology and abnormal astrocytic putrescine/GABA metabolism. Experimental inhibition of astrocytic ODC1 reduces aberrant GABA production, improves memory deficits, and can markedly reduce hippocampal Aβ plaque burden in APP/PS1 models.
