TOPflash is not an endogenous gene or protein but rather a reporter construct that is widely used in experimental studies to assess the transcriptional activity of the canonical Wnt/β-catenin signaling pathway.
By comparing the activity of TOPflash with that of a control reporter (such as FOPflash, which contains mutated TCF binding sites), investigators assess the specific contribution of Wnt/β-catenin signaling in cancer cell lines or experimental models.

Instead, the prognostic relevance lies in the fact that high activity of the canonical Wnt pathway—often inferred from assays using TOPflash—has been associated with more aggressive cancer phenotypes and poorer prognoses in several tumor types.
