RNA Polymerase I - DNA-Directed RNA Polymerase I

Type: Nuclear DNA-dependent RNA polymerase / ribosomal RNA transcription complex

Function: RNA polymerase I transcribes the 47S precursor ribosomal RNA, which is processed to generate 18S, 5.8S, and 28S rRNAs. Pol I activity is concentrated in the nucleolus and is a major regulator of ribosome biogenesis, protein-synthesis capacity, cell growth, and proliferation.

Cancer: ↑ Frequently hyperactivated in cancer. Increased Pol I-dependent rRNA transcription supports elevated ribosome biogenesis and protein synthesis required for rapid tumor-cell growth. Oncogenic pathways including MYC can stimulate Pol I transcription, while tumor suppressors including TP53 and RB normally restrain ribosome biogenesis. Pharmacological inhibition of Pol I can induce nucleolar stress and suppress tumor growth.
