Type: Nuclear DNA-dependent RNA polymerase / transcription complex

Function: RNA polymerase II transcribes essentially all protein-coding genes as precursor mRNAs as well as numerous non-coding RNAs. Its transcriptional cycle is regulated by general transcription factors, Mediator, chromatin regulators, and phosphorylation of the C-terminal domain of its largest subunit, POLR2A, particularly through transcription-associated kinases including CDK7, CDK9, and CDK12.

Cancer: ↕ Dysregulated / transcriptionally dependent. Cancer cells frequently become highly dependent on RNA polymerase II-mediated transcription to maintain expression of oncogenes, survival proteins, and short-lived transcriptional programs. Direct or indirect inhibition of Pol II transcription, including inhibition of CDK7 or CDK9, can preferentially suppress transcriptionally addicted cancer cells. A universal increase in Pol II expression itself is not characteristic of all cancers.
