Type: Nuclear DNA-dependent RNA polymerase / non-coding RNA transcription complex

Function: RNA polymerase III transcribes several abundant small non-coding RNAs required for protein synthesis and cellular growth, including transfer RNAs (tRNAs), 5S ribosomal RNA, and 7SL RNA. Pol III transcription is regulated by transcription factors including TFIIIB and TFIIIC and is closely coupled to cellular growth and nutrient signaling.

Cancer: ↑ Frequently increased or hyperactivated in transformed cells. Elevated Pol III transcription increases production of tRNAs, 5S rRNA, and other small RNAs required to sustain high rates of protein synthesis and tumor-cell proliferation. Oncogenic factors including MYC stimulate Pol III transcription, whereas tumor suppressors including TP53 and RB normally repress the pathway.
