| Ceramide - Bioactive Sphingolipid Type: Sphingolipid / bioactive lipid mediator / metabolic intermediate Function: Ceramide is a central intermediate in sphingolipid metabolism and a bioactive signaling lipid generated through de novo synthesis, sphingomyelin hydrolysis, and salvage pathways. Ceramide regulates apoptosis, cell-cycle arrest, autophagy, mitophagy, necroptosis, oxidative stress, mitochondrial function, membrane organization, and inflammatory signaling. Individual ceramide species differ according to fatty-acyl chain length and can have distinct biological effects. Cancer: ↕ Context-dependent. Ceramide accumulation frequently promotes cancer-cell death through apoptosis, mitochondrial dysfunction, mitophagy, necroptosis, and cellular stress signaling. Many cancers reduce pro-death ceramide signaling by converting ceramide into sphingosine-1-phosphate, glucosylceramide, or sphingomyelin. However, selected ceramide species can support tumor-cell survival depending on chain length, cellular compartment, and cancer type. Alzheimer's Disease: ↑ Ceramide levels are frequently elevated in Alzheimer's disease and can increase early in disease progression. Elevated ceramide promotes amyloid-β accumulation, oxidative stress, lipid peroxidation, mitochondrial dysfunction, neuroinflammation, and neuronal apoptosis and is associated with cognitive decline and increased risk of Alzheimer's disease. |
